Bacterial Expression

GST-fusion protein purification.

Buffers:

10 x PBS(0,3 M NaCl) for 1 L.

1 x TBS (Wash Buffer).

Tris-HCl pH 7.5

Thrombin cleavage buffer.

TBS + 2.5 mM CaCl2

Protocol:

1) Resuspend bacteria from 500 ml culture in 40 ml PBS +DTT, PMSF, leupeptin, pepstatin.

2) Sonicate with a microtip at the maximal intensity (2 ways):

a) 30 x 1 second at10 seconds interval.

b) 5 x 30 seconds (be sure to avoid overheating).

3) Add 10% triton to a final concentration of 1 % and centrifuge 15 min. at 15 000 RPM in SS-34 or 20 min. at 10 000 RPM in HB-4 rotor.

4) Pour the supernatant and add 700 ul of 50% GST-Sepharose. Incubate for 20 minutes at room temperature with moderate shaking.

5) Wash four times the loaded resin with 20 volumes of PBS (0,3 M NaCl), one time with TBS and two times with Thrombin cleavage buffer. (It is also possible to do these steps on a Biorad column)

6) Add 12,5-25 units of thrombin per litre of culture and incubate for one hour at room temperature in enough buffer to mix the resin from time to time.

7) Collect the eluate and freeze immediately.

 

 

 

Removal of nucleic acids using protamine

1) Resuspend bacteria from 500 ml culture in 40 ml PBS +DTT, PMSF, leupeptin, pepstatin.

2) Sonicate with a microtip at the maximal intensity (2 ways):

a) 30 x 1 second at 10 seconds interval.

b) 5 x 30 seconds (be sure to avoid overheating).

3) Add 10% triton to a final concentration of 1 % and centrifuge 15 min. at 15 000 RPM in SS-34 or 20 min. at 10 000 RPM in HB-4 rotor.

4) Add 5.14 g NaCl to the supernatant (to a final concentration of 2 M), adjusting the volume to 45 ml if needed.

5) Add 5 ml of 100 mg/ml protamine sulfate from salmon sperm (solubilised by heating) to a final concentration of 10 mg/ml.

6) Dialyse overnight in Spectrapor 12-14 000mw tube against 50 mM Tris-Hcl pH 7,5, 0,3 M NaCl , 1 mM DTT and 0,1 mM PMSF.

7) Pour the supernatant and add 700 ul of 50% GST-Sepharose. Incubate for 20 minutes at room temperature with moderate shaking.

8) Wash four times the loaded resin with 20 volumes of PBS (0,3 M NaCl), one time with TBS and two times with thrombin cleavage buffer. (It is also possible to do these steps on a Biorad disposable column)

9) Add 12.5 to 25 units of thrombin per litre of culture and incubate for one hour at room temperature in enough buffer to allow easy resuspension the resin from time to time.

10) Collect the eluate and freeze immediately.

 

 

Protocole de purification de HMG Boxes 1 à 3 de l’UBF par HPLC

Après incubation avec protamine, dialyse, passage sur G-Sepharose et digestion à la thrombine, on acidifie l’extrait avec HCl (conc) jusqu’à pH. 2 (environ 3,5 ul/ml de solution).

(Lorsque GST-Box3 est sur G-Sepharose, on fait un lavage avec 5 vol de résine de ATP (5 mM) pour se débarasser de GroES)

(La dialyse de GST-Box1 doit être la plus courte possible (2 x 1,5h) car il y a dégradation importante)

On centrifuge l’extrait acidifié qui est prêt à être chargé sur une colonne C-4 semi-préparative, sur HPLC (les programmes 20 et 16 sur le HPLC Biorad, servent au lavage des colonnes et de programme gabarit pour la purification des boîtes HMG)

Bouteille A: eau dégazéifiée + 0,05% TFA

Bouteille B: acétonitrile (Burdick and Jackson) + 0,05% TFA

Le débit pour chaque programme est de 1,5 ml/min.

Les boîtes HMG 1,2 et 3 sont dans le dernier pic, qui est moins pointu, on repasse une seconde fois les boîtes 1 et 3. Lors de ce passage, on peut augmenter le pourcentage d’acétonitrile de 1 ou 2% pour collecter la protéine dans un volume plus petit.(on met une légère pente dans l’élution pour contrecarrer l’imprécision des pompes). On ajoute 2 x la molarité des sites chargés de NaCl pour se débarasser du TFA lors de la lyophilisation finale.
